Biotin, also known as vitamin H or B7, has the chemical formula C10H16N2O3S (Biotin; Coenzyme R, Biopeiderm), is a water-soluble B-complex vitamin which is composed of an ureido (tetrahydroimidizalone) ring fused with a tetrahydrothiophene ring. A valeric acid substituent is attached to one of the carbon atoms of the tetrahydrothiophene ring. Biotin is a cofactor in the metabolism of fatty acids and leucine, and in gluconeogenesis.

Biotin is a B vitamin that’s needed for the formation of fatty acids and glucose, which are essential for the production of energy. It also helps with the metabolism of carbohydrates, fats and proteins.

Biotin is necessary for cell growth, the production of fatty acids, and the metabolism of fats and amino acids. It plays a role in the Citric acid cycle, which is the process by which biochemical energy is generated during aerobic respiration. Biotin not only assists in various metabolic reactions, but also helps to transfer carbon dioxide. Biotin is also helpful in maintaining a steady blood sugar level. Biotin is often recommended for strengthening hair and nails. Consequently, it is found in many cosmetic and health products for the hair and skin.

Deficiency is extremely rare, as intestinal bacteria generally produce an excess of the body’s daily requirement. For that reason, statutory agencies in many countries (e.g., the Australian Department of Health and Aging) do not prescribe a recommended daily intake.

Biotin deficiency isn’t common, unless you frequently eat a lot of raw egg white, which contains a protein that blocks the absorption of biotin. Genetic disorder of biotin deficiency, infant seborrheic dermatitis, surgical removal of the stomach, and excessive alcohol consumption may increase a person’s requirement for biotin.

Biotin deficiency may lead to skin rash, hair loss, high cholesterol and heart problems.

Dietary
Biotin is widely distributed in a variety of foods, but most often at low concentrations. Estimates are that the typical U.S. diet provides roughly 40 mcg/day. There are only a couple of foods which contain biotin in large amounts, including royal jelly and brewer’s yeast. The most important natural sources of biotin in human nutrition are milk, liver, egg (egg yolk), and some vegetables.Biotin is found naturally in food. Good dietary sources of biotin include brewer’s yeast, nutritional yeast, cauliflower, salmon, bananas, carrots,  sardines, legumes and mushrooms.

The most important natural sources in feeding nonruminant animals are oilseed meals, alfalfa, and dried yeasts. It is important to note that the biotin content of food varies and can be influenced by factors such as plant variety, season, and yield (endosperm-to-pericarp ratio).

Adequate intake are determined for nutrients when there is insufficient scientific evidence to establish a Recommended Dietary Allowance (RDA). These values are set as goals for individuals to support adequate nutritional status. NOTE: U.S. Food and supplement labels show 30 mcg of biotin as providing only 10% DV (Daily Value) because DVs are based on older and in some instances outdated RDAs for nutrients. Thus, the DV for biotin is 300 mcg even though there is now consensus that 30 mcg is adequate. There is no current Tolerable Upper Limit (UL) set for biotin as research has indicated that high levels of intake by humans has no detrimental effects.

Bioavailability
Studies on the bioavailability of biotin have been conducted in rats and in chicks. From these studies, it was concluded that biotin bioavailability may be low or variable depending on the type of food being consumed, but in general, approximately half of the biotin in most foods is considered to be biologically available. The biotin present in corn is readily available; however, most grain have about a 20-40% bioavailability of biotin .

A possible explanation for the wide variability in biotin bioavailability is that it is due to ability of an organism to break various biotin-protein bonds from food. Whether an organism has an enzyme with the ability to break that bond will determine the bioavailability of biotin from the foodstuff.

Factors that Affect Biotin Requirements
The frequency of marginal biotin status is not known, but the incidence of low circulating biotin levels in alcoholics has been found to be much greater than in the general population. Also, relatively low levels of biotin have been reported in the urine or plasma of patients who have had partial gastrectomy or who have other causes of achlorhydria, burn patients, epileptics, elderly individuals, and athletes. Pregnancy and lactation may be associated with an increased demand for biotin. In pregnancy, this may be due to a possible acceleration of biotin catabolism, whereas in lactation, the higher demand has yet to be elucidated. Recent studies have shown that marginal biotin deficiency can be present in human gestation, as evidenced by increased urinary excretion of 3-hydroxyisovaleric acid, decreased urinary excretion of biotin and bisnorbiotin, and decreased plasma concentration of biotin. Additionally, smoking may further accelerate biotin catabolism in women.

Medicinal Uses:

Hair Problems
Biotin supplements are often recommended as a natural product to counteract the problem of hair loss in both children and adults. There are, however, no studies that show any benefit in any case where the subject is not actually biotin deficient. The signs and symptoms of biotin deficiency include hair loss which progresses in severity to include loss of eye lashes and eye brows in severely deficient subjects. Some shampoos are available that contain biotin, but it is doubtful whether they would have any useful effect, as biotin is not absorbed well through the skin.

Cradle cap (seborrheic dermatitis)
Children with a rare inherited metabolic disorder called phenylketonuria (PKU; in which one is unable to break down the amino acid phenylalanine) often develop skin conditions such as eczema and seborrheic dermatitis in areas of the body other than the scalp. The scaly skin changes that occur in people with PKU may be related to poor ability to use biotin. Increasing dietary biotin has been known to improve seborrheic dermatitis[citation needed] in these cases.

Diabetes
People with type 2 diabetes often have low levels of biotin. Biotin may be involved in the synthesis and release of insulin. Preliminary studies in both animals and people suggest that biotin may help improve blood glucose control in those with diabetes, particularly type 2 diabetes. Specifically, biotin doses in excess of nutritional requirements lower postprandial glucose and improve glucose tolerance.

Deficiency:-
Biotin deficiency is relatively rare and mild, and can be addressed with supplementation. Such deficiency can be caused by the excessive consumption of raw egg whites, which contain high levels of the protein avidin, which binds biotin strongly. Avidin is deactivated by cooking, while the biotin remains intact.

Biotinidase deficiency is not due to inadequate biotin, but rather to a deficiency in the enzymes which process it.

Signs of Biotin Deficiency: In general, appetite and growth are decreased. Dermatologic symptoms include dermatitis, alopecia, and achromotrichia (absence or loss of pigment in the hair). Perosis (a shortening and thickening of bones) is seen in the skeleton. FLKS (fatty liver and kidney syndrome) and hepatic steatosis also can occur.

Toxicity
Animal studies have indicated few, if any, effects due to toxic doses of biotin. This may provide evidence that both animals and humans may tolerate doses of at least an order of magnitude greater than each of their nutritional requirements. There are no reported cases of adverse effects from receiving high doses of the vitamin, particularly when used in the treatment of metabolic disorders causing sebhorrheic dermatitis in infants.

Side Effects and Safety Concerns:-
The safety of biotin supplements in pregnant or nursing women, children or people with liver or kidney disease isn’t known.

People with a history of seizures shouldn’t use biotin unless under the supervision of a qualified health practitioner

Definition: Alpha lipoic acid is a fatty acid found naturally inside every cell in the body. It’s needed by the body to produce the energy for our body’s normal functions. Alpha lipoic acid converts glucose (blood sugar) into energ..

Alpha lipoic acid is also an antioxidant, a substance that neutralizes potentially harmful chemicals called free radicals. What makes alpha lipoic acid unique is that it functions in water and fat, unlike the more common antioxidants vitamins C and E, and it appears to be able to recycle antioxidants such as vitamin C and glutathione after they have been used up. Glutathione is an important antioxidant that helps the body eliminate potentially harmful substances. Alpha lipoic acid increases the formation of glutathione.

Alpha lipoic acid is made by the body and can be found in very small amounts in foods such as spinach, broccoli, peas, Brewer’s yeast, brussel sprouts, rice bran, and organ meats. Alpha lipoic acid supplements are available in capsule form at health food stores, some drugstores, and online. For maximum absorption, the supplements should be taken on an empty stomach.

It is an antioxidant that is manufactured in the human body. Antioxidants are substances that work by attacking “free radicals,” waste products created when the body turns food into energy. There are also many sources of free radicals in the environment such as ultraviolet rays, radiation, and toxic chemicals in cigarette smoke, car exhaust, and pesticides. Free radicals cause harmful chemical reactions that can damage cells in the body, making it harder for the body to fight off infections. As a result a person becomes more susceptible to long term diseases such as diabetes and liver damage.

Uses:

*Peripheral Neuropathy

Peripheral neuropathy can be caused by injury, nutritional deficiencies, chemotherapy or by conditions such as diabetes, Lyme disease, alcoholism, shingles, thyroid disease, and kidney failure. Symptoms can include pain, burning, numbness, tingling, weakness, and itching.


Alpha lipoic acid is thought to work as an antioxidant in both water and fatty tissue, enabling it to enter all parts of the nerve cell and protect it from damage.

Preliminary studies suggest that alpha lipoic acid may help. In one of the largest studies on the use of alpha lipoic acid, 181 people took 600 mg, 1200 mg or 1800 mg of alpha lipoic acid a day or a placebo. After 5 weeks, alpha lipoic acid improved symptoms. The dose that was best tolerated while still providing benefit was 600 mg once daily.

*General

Alpha-lipoic acid works together with other antioxidants such as vitamins C and E. It is important for growth, helps to prevent cell damage, and helps the body rid itself of harmful substances

*Diabetes

Several studies suggest that treatment with ALA may help reduce pain, burning, itching, tingling, and numbness in people who have nerve damage (called peripheral neuropathy) caused by diabetes. Alpha-lipoic acid has been used for years for this purpose in Europe. Other studies have shown that alpha-lipoic acid speeds the removal of glucose (sugar) from the blood of people with diabetes and that this antioxidant may prevent kidney damage associated with diabetes in animals.

*Liver Disease

Alpha-lipoic acid may prove useful in the treatment of chronic hepatitis because it relieves stress on the liver and helps rid the body of toxins. There have been several case reports of use of alpha-lipoic acid in combination with silymarin (milk thistle) and selenium (a substance with liver-protecting and antioxidant properties) to help treat hepatitis C (a serious type of hepatitis contracted from blood and bodily fluids that does not have an adequate cure or treatment).

It has also been used in conjunction with silymarin to treat Amanita poisoning. Amanita is a highly poisonous mushroom that causes liver damage.

*Brain Function and Stroke

Because alpha-lipoic acid can pass easily into the brain, it has protective effects on brain and nerve tissue and shows promise as a treatment for stroke and other brain disorders involving free radical damage. Animals treated with alpha-lipoic acid, for example, suffered less brain damage and had a four times greater survival rate after a stroke than the animals who did not receive this supplement. While animal studies are encouraging, more research is needed to understand whether this benefit applies to people as well.

 

Alpha lipoic acid can cross the blood-brain barrier, a wall of tiny vessels and structural cells, and pass easily into the brain. It is thought to protect brain and nerve tissue by preventing free radical damage.

*Age-Related Conditions

As an antioxidant, alpha lipoic acid can neutralize free radicals which can damage cells. Free radical damage is thought to contribute to aging and chronic illness.

*Other Conditions

Additional conditions for which alpha-lipoic acid may prove useful include heart failure, human immunodeficiency virus (HIV), cataracts, and glaucoma. More research is underway in these areas.

Alpha lipoic acid has also been suggested for cataracts, multiple sclerosis, burning mouth syndrome, Alzheimer’s disease and stroke, but large, well-designed studies are needed to see if it’s effective for these conditions.

Dietary Sources

Good food sources of alpha-lipoic acid include spinach, broccoli, beef, yeast (particularly Brewer’s yeast), and certain organ meats (such as the kidney and heart.)

Available Forms
Alpha-lipoic acid supplements are available in capsule form.

How to Take It
*Pediatric

There are no known scientific reports on the pediatric use of alpha-lipoic acid. Therefore, it is not currently recommended for children.

*Adult

Alpha-lipoic acid can be purchased in dosages ranging 30 mg to 100 mg tablets. Currently there are no established recommended doses for supplementation. For general antioxidant support, the recommended dose of ALA is 20 mg to 50 mg per day.

Manufacturers of alpha-lipoic acid suggest one or two 50-mg capsules daily as a dietary supplement.

Studies that have been successful in improving nerve function in diabetics have used 600 mg of alpha-lipoic acid per day in divided doses.

Precautions:
Because of the potential for side effects and interactions with medications, dietary supplements should be taken only under the supervision of a knowledgeable healthcare provider. This is especially true for those who are pregnant or breastfeeding.

Skin rash has been reported rarely from alpha-lipoic acid.

Finally, because alpha-lipoic acid has been associated with improved blood sugar control, people with diabetes should follow their blood sugar levels carefully when taking this supplement in order to avoid hypoglycemia (low blood sugar). Your doctor may decide that a reduction in dosage of insulin or oral blood sugar-lowering drugs is needed if you are taking this supplement.

Side Effects
Side effects of alpha lipoic acid may include headache, tingling or a “pins and needles” sensation, skin rash, or muscle cramps.

There have been a few reports in Japan of a rare condition called insulin autoimmune syndrome in people using alpha lipoic acid. The condition causes hypoglycemia and antibodies directed against the body’s own insulin without previous insulin therapy.

The safety of alpha lipoic acid in pregnant or nursing women, children, or people with kidney or liver disease is unknown.

Possible Drug Interactions:
Alpha lipoic acid may improve blood sugar control, so people with diabetes who are taking medication to lower blood sugar, such as metformin (Glucophage), glyburide (DiaBeta, Glynase), should only take alpha lipoic acid under the supervision of a qualified health professional and have their blood sugar levels carefully monitored.

Animal studies indicate that alpha lipoic acid may alter thyroid hormone levels, so it could theoretically have the same effect in humans. People taking thyroid medications such as levothyroxine should be monitored by their healthcare provider.

If you are currently being treated with any of the following medications, you should not use alpha-lipoic acid without first talking to your healthcare provider.

Amikacin and Gentamicin

In an animal study, alpha-lipoic acid supplements reduced side effects, particularly toxicity to the ear, associated with these antibiotics. Additional studies are needed to confirm these effects in people.

Cisplatin and Cyclophosphamide

The use of alpha-lipoic acid supplements in animals protected against toxic side effects associated with these medications.

Thyroid-regulating Medications, Levothyroxine

Rats given alpha-lipoic acid supplements had altered thyroid hormone function, but improved cholesterol levels. Blood hormone levels and thyroid function tests should be monitored closely in people taking thyroid hormones who are also taking alpha-lipoic acid.

Attempts to prevent cardiovascular disease are more effective when they remove and prevent causes, and they often take the form of modifying risk factors. Krill oil, like fish oil, contains omega-3 fats such as eicosapentanoic acid (EPA) and docosahexanoic acid (DHA). However, in fish oil, these omega-3 fats are found in the triglyceride form. In krill oil, they are found in a double chain phospholipid structure. The fats in human cell walls are in the phospholipid form.

The phospholipid structure of the EPA and DHA in krill oil makes them much more absorbable. Krill oil also contains vitamin E, vitamin A, vitamin D and canthaxanthin, which is a potent anti-oxidant.

The anti-oxidant potency of krill oil is, in terms of ORAC (Oxygen Radical Absorptance Capacity) values, 48 times more potent than fish oil.

The astaxanthin found in krill oil provides also excellent protection against ultraviolet light and UV-induced skin damage.

Found in milk and yogurt, vaccenic acid — a naturally occurring trans fat — lowered cholesterol in rats, a Canadian study finds.

Trans fats: We’ve been told that they’re worse for our hearts than saturated animal fats. Now, as consumers increasingly turn to food that’s trans-fat-free and manufacturers pull them from more and more processed foods, comes a twist. Some trans fats, ones that exist naturally, may be good for you.

In a 4-month study at the University of Alberta presented in March at a scientific meeting, obese rats fed a diet enriched with vaccenic acid — a naturally occurring trans fat found in milk and yogurt — had significant reductions in total cholesterol, LDL (or “bad”) cholesterol and triglycerides.

The researchers reported that a key benefit of vaccenic acid is its ability to reduce the production of chylomicrons — small particles of fat, protein and cholesterol formed in the gut that transport fats to various tissues of the body.

Like humans, obese rats produce too many chylomicrons, which raises lipids in the bloodstream. After 16 weeks of consuming vaccenic acid-enriched chow, however, the levels of chylomicrons dropped by more than half.

It’s not clear what this finding means for humans. First, the study was done in rats — the researchers say they’re planning some human clinical trials with vaccenic acid supplementation. Second, because the study diets were supplemented with vaccenic acid, the amounts the rats ate relative to their body weight was more than we would naturally eat in our usual diet.

The study is in line with other reports that natural trans fats have different effects on the body than the industrially created ones.

Most of the trans fats we eat — by far — come from partially hydrogenated vegetable oils, produced from liquid oils by industrial processing to create a firmer fat. Others occur naturally in milk products, formed in the rumen (or first stomach) of ruminant animals such as cows, goats, sheep and yaks when they’re fed a grass-rich diet.

Several studies of large populations have looked at the link between trans fatty acid intake and risk of developing atherosclerosis, and all have shown that the risk goes up only with the intake of “industrial” trans fatty acids, not the natural ones. Several clinical trials — in which people were fed special diets for weeks or months — have shown that manufactured trans fats raise LDL cholesterol levels to the same degree as saturated fats, and also lead to lower levels of the good, or HDL, cholesterol. It’s been estimated that it takes only about 12 grams of manmade trans fats to see this effect.

Trans-fat-free foods are big business, and today the majority of margarines, cookies, snack cakes and chips are devoid of the stuff. The change was fueled by the fact that, two years ago, it became law that food labels disclose industrial trans fat content.

Even if all the partially hydrogenated vegetable oil disappeared from our foods, we’d still consume about 1% to 5% of our calories from naturally occurring trans fatty acids, mostly vaccenic acid.

At this point, it’s not known how much vaccenic acid we’d need to consume to reap benefits. But in the meantime, anyone wanting to increase their natural-trans-fat intake might want to develop a taste for exotic cheese.

A study published in February in the Journal of Agriculture and Food Chemistry reported that yak cheese, from animals grazing in alpine grassland, contains more than four times the vaccenic acid of conventional cheddar cheese from grain-fed dairy cows. (The study didn’t investigate the levels in cheese from grass-fed cattle.) It also contains three times more beneficial omega-3 fatty acids. The authors conclude that a daily serving of 3 ounces of yak cheese might promote health.

Yak cheese isn’t easy to find — but the bottom line seems to be that the fatty acid composition of milk, cheese and yogurt from grass-fed animals may be more healthful than we knew — and perhaps, when the clinical trials are done, vaccenic acid-rich milkfat may join the ranks of other healthful fats along with those found in fish oil and nuts.
